Gliclazide
Indications/Uses
Type II DM (non-insulin dependent diabetes) in adults.
Dosage/Direction for Use
1 tab once daily, increased if necessary up to 4 tab as a single dose.
Administration
Should be taken with food: Swallow whole, do not chew or crush.
Contraindications
Hypersensitivity. Not to be used in juvenile onset diabetes; diabetes complicated by ketosis or acidosis; diabetics undergoing surgery, after severe trauma or during infections; diabetic precoma & coma; severe renal or hepatic insufficiency, porphyria, hyperthyroidism; insulin-dependent diabetes (type 1) & patients taking medicines to treat fungal infections. Galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption syndrome. Pregnancy & lactation.
Special Precautions
Increased CV mortality. Capable of producing moderate to severe hypoglycemia in patients controlled by diet alone; in case of accidental overdose; deficient calorie or glucose intake. Dosage adjustments for mild symptoms of hypoglycemia (sweating, pallor, hunger pangs, tachycardia, & sensation of malaise). Loss of control may occur for patients stabilized on diabetic regimen exposed to stress eg, fever, trauma, injection or surgery. Hepatic &/or renal impairment. May impair ability to drive or operate machinery. Pregnancy & lactation.
Adverse Reactions
GI disturbances eg, nausea, vomiting, heartburn, anorexia, diarrhea & metallic taste; increased appetite & wt gain; skin rashes, pruritus, photosensitivity.
Drug Interactions
May potentiate hypoglycemic effect w/ phenylbutazone, salicylates, sulfonamides, coumarin derivatives, MAOIs, β-adrenergic blockers, ACE inhibitors, tetracycline compd, chloramphenicol, clofibrate, disopyramide, oral miconazole, cimetidine, oral antidiabetics, GLP-1 receptor agonists or insulin, H2-receptor antagonists & medicines containing alcohol. May diminish hypoglycemic action w/ corticosteroids, OCs, thiazide diuretics, phenothiazines derivative, thyroid hormones & abuse of laxatives. May reduce blood clotting effects of warfarin.
